Salmonella species are recognized as major zoonotic pathogens for human and animals. In many countries, Salmonella is the leading cause of food-borne outbreaks and infections. In order to minimize this risk for consumers, microbiological controls of food is being applied. Therefore there is a need of reliable and rapid test, which should be used internationally to indicate the presence of absence of these pathogens as these bacteria are important for agricultural and food industry. PCR is merging as the Gold standard for many micro organisms. Therefore PCR method can be applied for Salmonella.
Targetgene: highly conserved gene region of salmonella
Kinds of probes: direct detection in tissue, food, water, fodder, blood, feaces. This kit can be used to detect the presence of Salmonella sp. in samples like chicken /sheep/cow/pig/goat meat as well as feaces after first selective enrichment step. This first selective enrichment step can be a short preenrichment in buffered peptone water (BPW) (5-6 hours) followed by selective enrichment on modified semisolid Rappaport-Vassiliadis (MRSV) for feacal samples. MRSV is better than other as it can be used to detect motile salmonellae and is less appropriate for the detection of nonmotile salmonellae.
One can isolate the DNA from the sample through commercial available kits or in house methods.
Validation: checked for cross reactions with related and nonrelated micro organisms. It is screened for other gene targets of Salmonella for the specificity.
This is checked for specificity with different strains of Samonellaes like Salmonella enteritidis, S. Typhimurium, Typhi, Heidelberg Dublin etc. (around 43 strains), therfefore the kit is specific to detect wide range of Salmonella serotype including all subspecies.
Detection limit: 5 to 50 Salmonella CFU or 10 genomic copies of purified Salmonella DNA per reaction
Contents of the kits: Nucleotides, polymerase, buffers, PCR master mix, loading dye, positive control, negative control as well as molecular marker along with the manual.
They are ready to use form as there are only 2 steps to do for Thermocycling.
