Mycoplasma pneumoniae is a respiratory pathogen, which causes around 10-20% of all community acquired pneumoniae in human beings.It is very difficult to make the differentiate diagnosis between the patients suffering from Chlamydia pneumoniae and Mycoplasma pneumoniae, therefore there is a need of laboratory diagnosis. This laboratory diagnosis can be done through culture methods (which takes a longtime as Mycoplasma goes slowly and needs special media), serological methods like ELISA as well as FAT, which are prone to cross reactions, therefore the PCR is merging as a goldstandard for detection of micro organisms in medicine as this is highly sensitive and specific also. Moreover PCR needs a little material to conduct the test.
Target gene: highly conserved region of repetitive gene of Mycoplasma pneumoniae. Around 8% genome of Mycoplasma pneumoniae is made of repetitive DNA elements.
Kinds of samples: Throat swabs, respiratory system swabs, colonies grown on cultures, reproductive tract swabs, blood samples, tissue samples and other body fluid samples. This kit needs isolated DNA, which can be isolated using commercial available kits or home made methods.
Components of the kit: polymerase, forward and backward primers, nucleotides, buffers, PCR master mix, loading dye, gene ruler (100 bp), positive control, negative control as well as manual.
Validations: It has been tested for cross reactions with related and non related micror organisms like Mycoplasma genitalium, Chlaymdia trachomatis, Chlaymdia pneumoniae, Mycoplasma hominis, Ureaplasma urealyticum, Legionella pneumoniae, Styphococcus aereus, Bordetella pertusis, CMV, RSV and many more. It is tested for serial dilutions.
